Wales number eight Toby Faletau will fly home from the Australia tour after breaking his hand during Saturday's first Test defeat.
Caretaker coach Rob Howley has called up Melbourne Rebels' Gareth Delve as Faletau's replacement. The former Gloucester captain won the last of his 11 caps in 2010.
Wales warm up for the second Test in Melbourne with a match against the Brumbies in Canberra on Tuesday, though lock Luke Charteris is the sole survivor from the 27-19 Brisbane defeat.
Second row Alun-Wyn Jones will captain the team, taking over from the rested Sam Warburton, while highly-rated 19-year-old Harry Robinson will start on the wing.
"We are a game down, but we still have all to play for in this Test series and Tuesday night is the perfect opportunity for this squad of players to move on," said Howley.
"We have one or two bumps and bruises, and we are expecting players to back-up after playing a tough Test match and go out there 72 hours later and play for their country."
