Australia take on the Welsh at the Principality Stadium in Cardiff and the hosts are missing several key players going into their opening autumn internationals through injury.
British and Irish Lions players Sam Warburton, Justin Tipuric, Ross Moriarty and Dan Lydiate are all out of action and that has limited the options head coach Warren Gatland has for the match.
Josh Navidi, who will make his home debut, and Aaron Shingler will be the two flankers for the Welsh whilst number eight Taulupe Faletau completes the back-row.
Many feel Wales will struggle to win the game but Hooper is taking nothing for granted and has warned his team-mates to expect a difficult challenge on Saturday afternoon.
He said: "We know they're going to be class, Wales has put out quality back-rows as long as I've been playing."
